Regina did know about Cora setting her up to be Queen with Snow falling from the horse already, but she was so young and so abused that she blamed Snow instead and has launched such a long vendetta that she may well have "forgotten" that little part of the narrative until it was brought up to her again. I also believe Regina didn't realize Cora had killed Snow's mother prior to this (although we all did).

As for Regina playing her mother, I think she wants to know what her mother's plan is, sort of like in Ledger Squared in Blood.

That makes sense. I seriously hope the show is imitating Ledger Squared in Blood, that's a really good fic!

If they aren't purposely paralleling Emma and Regina's arcs, they are stumbling like (lucky) drunks into it.

Those parallels have to be on purpose, nobody's that dense!!
